Self-Nanoemulsifying Drug Delivery Systems for Improving Oral Delivery of Poorly Water-Soluble Drugs: Formulation Strategies, Characterization, Optimization and Therapeutic Applications

Poor aqueous solubility is a major challenge in oral drug delivery and can limit dissolution and absorption. Self-nanoemulsifying drug delivery systems (SNEDDS) are lipid-based formulations that spontaneously form fine nanoemulsions in aqueous gastrointestinal fluids, improving drug solubilization and dissolution. This review discusses SNEDDS formulation principles, excipient selection, self-emulsification mechanisms, pseudo-ternary phase diagrams, Quality by Design, optimization, characterization, dissolution, precipitation, supersaturable and solid SNEDDS, therapeutic applications, limitations, and future prospects. Emphasis is placed on rational excipient selection, precipitation control, biorelevant evaluation, and in vivo validation for successful enhancement of oral drug delivery.
